miRNA-200a inhibits migration and invasion of nasopharyngeal carcinoma cells by down-regulating ZEB2

Abstract:Objective To investigate the effect and mechanism of miRNA-200a on the migration and invasion of nasopharyngeal carcinoma cells by down-regulating ZEB2.Methods qPCR was used to detect the expression of miRNA-200a in different nasopharyngeal carcinoma cell lines. Double luciferase reporter gene was used to detect the interactions between miRNA-200a and ZEB2. Scaling healing experiment and Transwell invasion assay were used to detect the effect of miRNA-200a on the migration and invasion of nasopharyngeal carcinoma cells. Western blot was used to detect the effect of miRNA-200a on the expression of ZEB2 protein.Results The expression level of miRNA-200a in CNE-1 cells was significantly higher than that in other nasopharyngeal carcinoma cell lines. Double-luciferase assay confirmed that miRNA-200a could regulate the expression and activity of ZEB2. Inhibition of miRNA-200a expression reduced the migration and invasion of nasopharyngeal carcinoma cells. The expression of ZEB2 was inhibited by the expression of miRNA-200a. Conclusion miRNA-200a can regulate the expression of ZEB2 and affect the migration and invasion of nasopharyngeal carcinoma cells.
