A quick method of isolating exosomes from mesenchymal stem cells using PEG 6000

Abstract:Objective To establish a method of exosomes extraction from bone marrow mesenchymal stem cells (BMSCs) using PEG 6000 via low-speed centrifugation. Methods The serum-free culture supernatant of BMSCs was harvested and centrifuged to remove cell debris before being filtered with a 0.22 μm micropore to clean up the smaller fragments and larger vesicles. The exosomes were purified using PEG 6000 method with low-speed centrifugation, followed by characterization using transmission electron microscopy. The magnetic bead technique and flow cytometry were employed to detect the expressions of CD9/D81/CD63/CD44. Matrigel was used to evaluate the effect of BMSCs-exo on tube formation of human umbilical vein endothelial cells. Results The BMSCs-exo were successfully separated by PEG 6000 combined with low-speed centrifugation. The BMSCs-exo were circular or oval, 100nm in diameter, and highly expressed in CD9/D81/CD63/CD44 molecules. The BMSCs-exo could promote tube formation of HUVECs in a concentration-dependent manner. Conclusion It is a simple and effective method to extract exosomes using PEG 6000 with low-speed centrifugation.
