Changes of Th17 and Treg cells and the cytokines levels in the peripheral blood of patients with acute pancreatitis

Abstract:ObjectiveTo study the changes of Th17 and Treg cells and the cytokines levels in the peripheral blood of patients with acute pancreatitis.MethodsNinety-two cases of acute pancreatitis treated in our hospital between August 2014 and August 2015 were selected and divided into three groups: MAP group (39 cases), MSAP group (29 cases),and SAP group (24 cases). Twenty-five healthy subjects were chosen as control group. The proportion of Treg and Th17 in peripheral blood from the four groups was detected by flow cytometry. mRNA of IL-17, IL-6, IL-10 and TGF-β in cells of peripheral blood was analyzed by real time PCR. Serum levels of IL-17, IL-6, IL-10 and TGF-β were detected by ELISA method.ResultsCompared with control group, the proportion of Treg cells and Treg/Th17 ratio in patients of MAP, MSAP and SAP groups significantly decreased (P<0.05), but the proportion of Th17 cells in patients of observation group was significantly increased (P<0.05). Compared with control group, IL-17 and IL-6 mRNA and protein levels in patients of MAP, MSAP and SAP groups were significantly increased (P<0.05), but IL-10 and TGF-β mRNA and protein levels in patients of observation group significantly decreased (P<0.05). ConclusionThere is imbalanced expression of Treg cells and Th17 cells and their cytokines in patients with acute pancreatitis, which is of clinical significance for study on the pathogenesis of acute pancreatitis.
