Production of NF-κB, VAP-1 and IL-6 inhibited by Ginkgo biloba extract in endothelial cells

Abstract:ObjectiveTo establish an oxidative stress model using HCAEs stimulated by activated platelets in vitro so as to explore whether Ginkgo biloba extract can inhibit the phosphorylation of NF- kappa B the production of interlukin-6 (IL-6) and vascular adhesion protein (VAP-1) in HCAECs, stimulated by activated platelets.MethodsActivated platelets were used to stimulate HCAECs to establish the oxidative stress model before, Ginkgo biloba extract (4, 40 or 400μg/ml) was used to act on them. Cell. culture supernatant was collected to measure the amount of IL-6 and VAP-1 via enzyme linked immune sorbent assay (ELISA), and total protein was extracted to examine the expression of NF-κB PP65 by Western Blot.ResultsThe results showed that the secretion of target molecules was increased significantly after HCAECs were stimulated by activated platelets. EGb at a therapeutic dose can inhibit the expression of p65, PP65 and the generation of IL-6,VAP-1, in a dose-dependent manner compared with activated platelets group (P<0.05).ConclusionEGb can attenuate the generation of IL-6,VAP-1 by inhibiting the expression of p65, PP65 significantly in HCAECs stimulated by activated platelets.
