Changes of serum angiotensin Ⅱ concentration in stress-induced cardiomyopathy animal models and effects of valsartan on stress-induced cardiomyopathy

Abstract:Objective To investigate the relationship between stress cardiomyopathy and angiotensin Ⅱ, and to observe the role of angiotensin Ⅱ receptor antagonists in the prevention and treatment of stress-induced cardiomyopathy. Methods Two-week-old female rabbits were randomly divided into the control group, the experimental group and the intervention group. Serum angiotensin Ⅱ concentration was detected by ELISA at 0, 7 and 14 d after operation, and the expression of AT1 and AT2 was measured by Western blot at 14 d. Myocardial tissue was observed by HE staining at 14 d. Results Angiotsnsin Ⅱ concentration in the experimental group and intervention group was more significantly increased than in control group at 0, 7 and 14 d after operation (P<0.05). At day 14, AT1 expression of the apex was significantly deceased in the experimental group compared with the control group (P<0.05), but was recovered by valsartan. Compared to the control group, AT1 expression of the apex was significantly increased in the experimental group and valsartan group(P<0.05). HE staining showed that the apical free wall of animals in the experimental group was significantly thinner than that of the control group (P<0.05), but was recovered by valsartan. Meanwhile, apical myocardial tissue in the experimental group showed typical cardiac stress cardiomyopathy. Conclusion Angiotensin Ⅱ may be involved in stress cardiomyopathy, and AT1 receptor antagonists can effectively prevent stress induced cardiomyopathy.
