The effect of LPS on the expression of TLR4/CD14/MD-2 complex in macrophage cells

Abstract:Objective To explore the effect of LPS on the expression of Toll-like receptor4(TLR4), cluster of differentiation 14 (CD14) and myeloid differentiation factor 2(MD-2) in RAW264.7 macrophages. Methods RAW264.7 macrophages were stimulated with 100 ng/ml of LPS or 1 000 ng/ml of LPS, the expressions of TLR4, CD14 and MD-2 mRNA at different time points were detected by RT-PCR. TNF-αlevels were measured by ELISA. Results In vitro treatment of RAW264.7 macrophages with 100 ng/ml of LPS decreased transcripts of TLR4 and increased transcription of CD14 and MD-2. Treatment with 1 000 ng/ml of LPS rescued transcription of TLR4 after 100 ng/ml LPS inhibition. Concentration of TNF-α had a higher level in high-dose LPS than low-dose LPS(P<0.05). Conclusion Low-dose LPS induced inhibition of TLR4 in vitro is reversed by high-dose LPS.
