Establishment of a novel miniature swine model of coronary artery stenosis for assessment of the effects of +Gz stresses

Abstract:Objective We intended to establish a type of animal model for studying maximal tolerated +Gz exposure of different coronary artery stenosis. Methods Of 25 Bama miniature swine, five swine underwent a sham operation as control group.Proximal left anterior descending branch (LAD) was ligated in 20 swine using silk suture by thoracoscope, to induce to different stenosis of LAD. +Gz exposure was performed after 1 week surgery, and then the maximal tolerated +Gz values were observed. Results ①Twenty miniature swine underwent LAD ligation, two swine suffered from ventricular fibrillation, and finally died 8 h and 48 h after surgery, respectively. Seven swine were in the mild stenosis group (36.8±7.4)%, 6 were in the moderate stenosis group (61.5±6.2)%, and 5 were in the severe stenosis group (81.9±8.6)%. The success rate of the surgery was 100%, and the success rate of the model construction was 90%.②The maximal tolerated+Gz values were: +(8.00±0.71)Gz in the sham-operated group, +(7.71±1.11)Gz in the mild stenosis group, +(6.00±0.89) Gz in the moderate stenosis group and + (5.20±0.84) Gz in the severe stenosis group. Compared with the sham-operated group, the maximal tolerated +Gz value of the moderate stenosis and severe stenosis groups both significantly decreased, but the mild group was not significantly different from the sham surgery control group. Conclusion The full-endoscopic operation could construct animal models of different coronary artery stenosis successfully with easier and faster recovery, and could allow enduring +Gz stress compared with the traditional models.
