Effect of dexmedetomidine on expression of neuronal nitric oxide synthase in the gray matter around the central canal in the spinal cord in a rat model of chronic neuropathic pain

Abstract:Objective To investigate the effect of dexmedetomidine on the pain threshold of hit and the expression of neuronal nitric oxide synthase (nNOS)in the gray matter around the central canal in the spinal cord in a rat model of chronic neuropathic pain. Methods Thirty-six male SD rats were randomly assigned into 3 groups: sham operation group (group Sham), chronic constrictive injury operation group (group CCI), CCI+dexmedetomidine group (group DEX). Chronic neuropathic pain was induced by chronic constrictive injury. The behavioral studies of rats were scored and thethermal withdrawal latency (TWL) was measured. The expression of nNOS in the gray matter around the central canal in the spinal cord was observed by immunohistochemistry. Results Compared with group Sham, TWL was significantly decreased and the expression of nNOS was up-regulated after operation in group CCI and group DEX(P<0.05). Compared with group CCI, TWL was significantly increased and the expression of nNOS was significantly down-regulated on day 7 and 14 after operation in group DEX(P<0.05). Conclusion Down-regulated nNOS in the gray matter around the central canal in the spinal cord is involved in the attenuation of neuropathic pain by DEX.
