Impact of dexmedetomidine on the deformability and antioxidative ability of human erythrocyte in vitro

Abstract:Objective We aimed to evaluate the impact of clinically relevant concentrations of dexmedetomidine on the deformability and antioxidative ability of human erythrocyte in vitro. Methods 5 ml intravenous blood samples taken from healthy adults were made into erythrocyte suspensions with 2%HCT in PBS buffer.Erythrocyte suspensions were divided into 4 groups:control group(group C);low concentration of dexmedetomidine group(group DL);middle concentration of dexmedetomidine(group DM);high concentration of dexmedetomidine(group DH).Group DL、DM、DH were pretreated with 3 different concentrations of dexmedetomidine (0.6ng/ml,1.8ng/ml or 5.4ng/ml) respectively.4 groups were placed into thermostatic incubator concussion 50rpm,with 37℃.After incubation for 1 h,the concentration of SOD and MDA of red blood cell and deformability of erythrocyte were measured respectively.Results After incubation for 1 h ,compared with group C(20.01±3.11)%,erythrocyte deformability index significantly increased in group DL(24.04±3.41)%,DM(25.03±3.45)% and DH (23.16±3.92)% (P<0.05).Compared with group C(14.885±0.510)U/mgHb,the activities of SOD significantly increased in group DM(15.830±0.503)U/mgHb(P<0.05).Compared with group C(4.182±0.236)nmol/mgHb,concentrations of MDA decreased in group DM (3.853±0.261)nmol/mgHb (P<0.05).Conclusion Dexmedetomidine treatment can improve deformability of human erythrocyte through enhancement of antioxidation of red blood cell.
