Induced differentiation in gastric cancer cell lines by site-selective 8-Br-cAMP
Abstract:Objective In order to observe the effect of site-selective8-Br-cAMP on the proliferation and differentiation in gastric cancer cell lines. Method MTT method was used to test the proliferation of cancer cells and the phenotypes of malignant cells were measured by Flowcytomytry(FCM). Result (1)8-Br-cAMP can inhibit the growth of gastric cancer cell line SGC7901, its IC50 value being at 40 μm; (2)Protein products of oncogene P21ras decreased by 30% after induced by 8-Br-cAMP, a tumor-associated antigen MGb2 also decreased by 51%. (3)When cancer cells were induced by 8-Br-cAMP(10 μmol/L)for 4 weeks, the formation rate of xenograft was obviously decreased in BALB/C nude mice compared with controlled group. However when induced with 40 μm 8-Br-cAMP, the cancer cells after induced can not produce the formation of xenograft. Conclusion 8-Br-cAMP may provided a new biological tool in experimental and therapeutic aspects in carcinoma.
