Observation on the Effect of Donepezil Combined with Rosuvastatin in the Treatment of Ischemic Stroke Complicated with Vascular Dementia

Abstract:Objective To explore the clinical efficacy of the combined application of donepezil and rosuvastatin in the treatment of patients with ischemic stroke complicated with vascular dementia.Methods A total of 124 patients with ischemic stroke complicated with vascular dementia who were diagnosed and treated in Pharmacy Department of Kaifeng People's Hospital from March 2021 to May 2024 were selected as the research subjects and randomly divided into the observation group(n=62)and the control group(n=62)according to the odd-even number method.The control group was treated with atorvastatin combined with donepezil,and the observation group was treated with rosuvastatin combined with donepezil.The dementia conditions before and after treatment were compared between the two groups and evaluated using the Hasegawa Dementia Scale(HDS)and the Mini-Mental State Examination(MMSE);the changes in blood lipid levels,including high-density lipoprotein cholesterol(HDL-C),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),and triglyceride(TG)levels;the changes in inflammatory factors,including C-reactive protein(CRP)and interleukin-1β(IL-1β)levels;the vascular endothelial function before and after treatment in the two groups was shown by the changes in endothelin-1(ET-1)and nitric oxide(NO)levels;and the adverse reactions occurred during the treatment process in the two groups were compared.Results After treatment,there was no significant difference in treatment efficacy between the observation group and the control group(P>0.05).The HDS,MMSE scores,and NO levels of the two groups were significantly higher than those before treatment,and the scores of the observation group were higher than those of the control group(P<0.05).The levels of CRP,IL-1β,and ET-1 in the two groups were lower than those before treatment,and the levels of CRP,IL-1β,and ET-1 in the observation group were lower than those in the control group(P<0.05).The levels of TG,TC,and LDL-C in the observation group were lower than those before treatment,and the level of HDL-C was higher than that before treatment(P<0.05).There was no significant change in the blood lipid levels of the control group compared with those before treatment(P>0.05).The levels of TG,TC,and LDL-C in the observation group were significantly lower than those in the control group,and the level of HDL-C was significantly higher(P<0.05).There was no significant difference in the total incidence of adverse reactions such as dizziness,diarrhea,sleep disorders,and joint pain between the two groups(P>0.05).Conclusion Donepezil combined with rosuvastatin has a prominent therapeutic effect in the treatment of ischemic stroke complicated with vascular dementia,which can reduce the inflammatory level of patients,reduce the damage of lipotoxicity to vascular endothelium,and improve cognitive ability.This regimen has good safety and is worthy of clinical promotion and use.
