Preparation Process of Total Alkaloids Liposomes from Suaeda salsa and Analysis of Their Antibacterial Activity

Abstract:Objective To optimize the preparation process of total alkaloid liposomes from Suaeda salsa and study their antibacterial activity.Methods The total alkaloid liposomes of Suaeda salsa were prepared using the thin film dispersion method.The encapsulation efficiency of the liposomes was used as an indicator.Based on a single factor experiment,the Box Behnken experimental design principle was used to investigate the effects of phospholipid to cholesterol molar ratio,phospholipid to drug mass ratio,hydration time and phospholipid concentration on encapsulation efficiency.Results The optimal prescription for the total alkaloid liposomes of Suaeda salsa was a phospholipid to cholesterol molar ratio of 6∶1,a phospholipid to drug mass ratio of 11∶1,a hydration time of 2 hours and a phospholipid mass concentration of 19 mg/mL.At this time,the encapsulation efficiency of the liposomes was 81.52%.In vitro release experiments showed a cumulative release rate of 63.10%within 24 hours.The total alkaloid liposomes of Suaeda salsa had antibacterial activity against Staphylococcus aureus and Bacillus subtilis,with minimum inhibitory concentrations of 500 and 700 μg/mL,respectively.Conclusion The optimized prescription process of this experiment is stable and feasible,and the total alkaloid liposomes of Suaeda salsa can prolong the drug's action time and have good antibacterial activity.
