Preparation of Metaplexis japonic Total Flavonoids-loaded Liposomes and Analysis of Their in Vitro Anti-tumor Activity

Abstract:Objective To prepare liposomes of total flavonoids from the Metaplexis japonic and analyze its anti-tumor activity in vitro.Methods The total flavonoid liposomes of Metaplexis japonic were prepared using the thin film dispersion method.The encapsulation efficiency was used as the evaluation index,and the lipid drug ratio,phosphate bile ratio,phosphate buffer solution dosage,and phospholipid mass concentration(PBS,pH=6.5)were the influencing factors.The Box-behnken response surface methodology was used to optimize the preparation process of the total flavonoid liposomes of Metaplexis japonic;the MTT assay was used to evaluate the inhibitory effect of the obtained liposomes on human liver cancer cell line HepG2.Results The optimal preparation process for total flavonoids liposomes from Metaplexis japonic was a lipid drug ratio of 8:1,phosphate bile ratio of 9:1,PBS dosage of 10 mL,and phospholipid mass concentration of 22 mg/mL,the encapsulation efficiency of 87.20%,and the cumulative release rate within 24 hours was 70.20%.MTT experiments showed that the liposomes had a more significant inhibitory effect on HepG2 cells.Conclusion The optimized preparation process of total flavonoids liposomes from Metaplexis japonica is feasible,and the liposomes have certain sustained release and anti-tumor activity in vitro.
