miR-25-3p Promotes Motor Function Recovery After Spinal Cord Injury in Rats by Targeting PTEN Through the AKT/mTOR Pathway

Abstract:Objective To investigate the effects of miR-25-3p on neuronal apoptosis and motor function in rats after spinal cord injury(SCI).Methods 103 male SD rats were selected and randomly divided into 4 groups:the sham operation group(Sham group),the spinal cord injury group(SCI group),the miR-25-3 p mimic injection group(agomiR group),and the miRNA negative control injection group(agoNC).The expression changes of miR-25-3p and PTEN before and after SCI were detected by real-time fluorescent quantitative PCR and protein immunoblotting;the expression changes of PTEN and AKT/mTOR pathway as well as apoptosis-related proteins Bcl-2,Bax and C-caspase3 before and after miR-25-3p mimics injection were also examined;the binding between miR-25-3p and PTEN was verified by dual luciferase reporter gene assay;the changes in motor function and the number of spinal motor neurons in each group of rats were evaluated by BBB score and Nissl staining.Results The apoptosis level of spinal cord tissue after SCI significantly increased,while the expression of miR-25-3p decreased and the expression of PTEN increased.At the same time,the expressions of p-AKT,p-mTOR and p-S6 significantly decreased.miR-25-3p can target and inhibit the expression of PTEN.Local injection of miR-25-3p mimics can significantly inhibit the apoptosis level of spinal cord tissue,increase the number of surviving motor neurons,and promote the recovery of motor function in rats.Conclusion After miR-25-3p targets PTEN,it inhibits neuronal apoptosis through the AKT/mTOR pathway,thereby promoting the recovery of motor function in rats.
