Effect of Norcantharidin on HGF-activated PI3K/AKT and NF-κB Signaling Pathway in Hypopharyngeal Carcinoma Cells

Abstract:Objective To observe the effects of norcantharidin(NCTD)on the PI3K/AKT and NF-κB pathways of hypopharyngeal cancer cells induced by hepatocyte growth factor(HGF),and to explore the possible mechanism by which norcantharidin inhibits the proliferation and invasion of hypopharyngeal cancer.Methods The hypopharyngeal cancer cells FaDu induced by HGF were treated with different concentrations of NCTD,and the changes in protein expression of the PI3K/AKT pathway and NF-κB pathway were detected by Western Blot assay.The activation of NF-κB was detected by immunofluorescence staining.Results(1)After NCTD treatment,the expression of p-c-met protein in each group of cells decreased.Compared with the HGF treatment group,the differences were statistically significant(P<0.01);(2)the protein expressions of p-PI3K and p-Akt in different concentrations of NCTD treatment groups decreased.Compared with the HGF treatment group,the differences were statistically significant(P<0.01);(3)the results of immunofluorescence staining revealed that after FaDu cells were treated with HGF,the fluorescence of NF-κB protein was mainly expressed in the nucleus.With the gradual increase of NCTD concentration,the fluorescence of NF-κB protein in the nucleus gradually weakened or even disappeared.Conclusion NCTD can inhibit the activation of the PI3K/AKT and NF-κB signaling pathways induced by HGF.
