The Anti-fatigue Effect of Shouzheng Drink as Tea Based on Network Pharmacology and Animal Experiments

Abstract:Objective This paper was aimed to study the anti-fatigue effect of shouzheng drink as tea through network pharmacology methods and animal experiments.Methods The active ingredients of shouzheng drink as tea were screened through traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)and traditional Chinese medicine integrated database(TCMID).Fatigue related targets were searched through the Genecards database.Venny 2.1 was used to obtain the targets of shouzheng drink as tea against fatigue,and the PPI network model was established through the STRING database.Gene ontology(GO)and kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis was performed on key targets through database for annotation,visualization and integrated discovery(DAVID),and Cytoscape 3.9.1 software was used to construct a network diagram of"potential active ingredients of shouzheng drink as tea-anti-fatigue related target-pathway".Molecular docking validation was performed by AutoDock Tools.Animal experiment was used to verify the anti-fatigue effect of shouzheng drink as tea.Results Shouzheng drink as tea included sixteen core ingredients related fatigue,such as quercetin,myristerol,kaempferol,and acted on 24 core targets,such as PRKACA,heat shock protein 90 alpha family class B member 1 gene(HSP90AB1),and adrenoceptor beta 2(ADRB2),which involved in cell division,response to calcium ion,enzyme binding,AMP-activated protein kinase(AMPK)signaling pathway,cyclic adenosine monophosphate(cAMP)signaling pathway,etc.The core components binded well to the core targets.Shouzheng drink as tea could prolong the weight-bearing swimming exhaustion time of mice and did not affect their body weight.Conclusion Shouzheng drink as tea might regulate various metabolic pathways such as glucose and lipid metabolism,energy metabolism by acting on PRKACA,ADBR2 and other targets,and further regulate AMPK and other signaling pathways to exert anti-fatigue effects.
