Mendelian Randomization Study on the Occurrence of Psoriasis Caused by Multiple Unhealthy Lifestyles

Abstract:Objective To explore the causal relationship between various unhealthy lifestyles,including obesity,smoking,drinking,and physical activity,and psoriasis through Mendelian randomization analysis.Methods From the data collected from genome-wide association studies(GWAS),single nucleotide polymorphisms(SNPs)strongly associated with unhealthy lifestyles were selected as instrumental variables.The main analysis used inverse variance weighting(IVW)to estimate causal effects,and mendelian candomization method based on egger(MR-Egger),weighted median method,and weighted median method were used as supplementary methods.To evaluate the robustness of the results,we also conducted sensitivity analysis,including Cochran's Q-test,MR Egger intercept,and leave-one-out method.Results Individuals with larger obesity related indicators were more prone to psoriasis,with BMI(OR=1.446,95%CI:1.327-1.574,P=2.43×10-17,WHR(OR=1.33,95%CI:1.18-1.498,P=2.75×10-6);smoking was associated with the risk of psoriasis,with Age Smk(OR=0.39,95%CI:0.191-0.798,P=0.00987457)starting smoking and Smklnit(OR=1.356,95%CI:1.233-1.491,P=3.51×10-10)regularly smoking.In addition,MR Egger intercept,Cochran's Q test,and leave-on-out did not observe the presence of horizontal pleiotropy and heterogeneity.Conclusion There is a causal relationship between obesity,smoking,and the occurrence of psoriasis.It is recommended to actively control obesity and reduce smoking to reduce the incidence of psoriasis.
