Effects of Curcumin on PI3K/AKT/mTOR Signaling Pathway in Myocardial Tissue of Type 2 Diabetes Rats

Abstract:Objective To investigate the protective effect and mechanism of curcumin on myocardial injury in type 2 diabetes rats.Methods Male SD rats were fed with high fat and high sugar diet for 4 weeks,and then intraperitoneal injection of STZ was used to establish the model of type 2 diabetes.Curcumin protection group was treated by intragastric administration of curcumin for 12 weeks.Fasting blood glucose and cardiac index of rats were detected;HE staining was used to observe the morphological changes of myocardial tissue;the SOD activity and MDA content in the myocardial tissue of rats in each group were measured using a reagent kit;Western Blot was used to detect the expression level of myocardial tissue related proteins in each group of rats.Results Compared with the control group,the heart index of the model group rats was significantly increased(P<0.05);myocardial tissue morphology was disturbed;SOD activity was significantly decreased(P<0.05);the MDA content was significantly increased(P<0.05);the levels of PI3k/β-actin,P-Akt/AKT and mTOR/β-actin were significantly decreased(P<0.05).Compared with the model group,the cardiac index of rats in the curcumin protected group was significantly reduced(P<0.05);myocardial tissue morphology was improved;SOD activity was significantly increased(P<0.05);the MDA content was significantly decreased(P<0.05);the levels of PI3k/β-actin,P-Akt/AKT,mTOR/β-actin were significantly increased(P<0.05).Conclusion Curcumin can improve the activity of PI3K/AKT/mTOR signaling pathway in myocardial tissue of diabetes rats through its own antioxidant activity,and can play a role in myocardial protection.
