Curcumin Alleviates Cerebral Ischemia-reperfusion Injury in Rats by Regulating Mitophagy

Abstract:Objective To investigate the regulatory effect of curcumin on mitophagy in rats with cerebral ischemia-reperfusion(MCAO/R)and its possible mechanism.Methods The MCAO/R model was prepared by wire plug method,and the degree of neurological impairment in rats was evaluated by Zea-Longa scoring method;dry and wet gravimetric methods were used to detect cerebral edema in rats in each group;the cerebral infarction volume of rats in each group was determined by 2,3,5 triphenyltetrazolium chloride(TTC)staining;the expression levels of ROS and ATP in each group were detected by the kit;qRT-PCR was used to explore the expression levels of P62,LC3 Ⅱ and TOM20 mRNA;enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of inflammatory cytokines IL-6,TNF-α,IL-1 β and IL-10;the expression of IL-6 and IL-10 in rats was detected by immunofluorescence.Results The results of Zea-Longa score,TTC staining and cerebral edema evaluation showed that compared with the MCAO/R group,the curcumin treatment group could significantly improve the neurological function of rats and reduce the volume of cerebral infarction and cerebral edema.The results of qRT-PCR showed that the expression of P62 mRNA and TOM20 mRNA decreased and the expression of LC3 Ⅱ increased in the curcumin treatment group.The results of ELISA experiments showed that the expressions of pro-inflammatory factors IL-6,TNF-α and IL-1 β were decreased and the expression of anti-inflammatory factors IL-10 was increased in the curcumin treatment group.Conclusion Curcumin inhibits the inflammatory response of SD rats after cerebral ischemia and reperfusion by activating the autophagy function of mitochondria,thereby promoting the recovery of neurological function after cerebral ischemia and reperfusion,and exerting its neuroprotective role.
