Study on Proliferation of Ovarian Serous Cancer Cells Regulated by Long Non-coding RNA LINC00641

Abstract：Objective To explore the expression of long non-coding RNA LINC00641(LINC00641)in ovarian serous carcino-ma and investigate its regulatory role in proliferation.Methods The GEPIA database was applied to predict the trend of LINC00641 expression in ovarian cancer.57 cases of ovarian serous carcinoma tissues were selected as the observation group,and 57 ovarian se-rous cystadenoma tissues were selected as the control group.Real-time quantitative fluorescence PCR(qRT-PCR)was used to detect the expression of LINC00641,and immunohistochemistry was used to assess Ki67 expression.The human ovarian serous carcinoma cell line SKOV3 and human ovarian surface epithelial cells HOSEpiC were selected.qRT-PCR was used to detect LINC00641 expres-sion.SKOV3 cells were transfected with LINC00641 plasmid to establish the OE-LINC00641 group,with a blank control group(NC).Cell proliferation activity was assessed using CCK-8.Results The GEPIA database showed a trend of downregulation of LINC00641 expression in ovarian cancer(P<0.05).The expression of LINC00641 in ovarian serous carcinoma tissues(1.325±0.115)was significantly lower than that in the control group(1.665±0.147)(t=6.24,P<0.05).LINC00641 showed significant differences in different lesion grades(1.31±0.11 vs 1.43±0.09)and maximum tumour diameters(1.25±0.11 vs 1.36±0.10)(P<0.05).There was a negative correlation between LINC00641 and Ki67 proliferation index in ovarian serous carcinoma(P<0.05).LINC00641 expression was lower in SKOV3(1.33±0.13)than that in HOSEpiC(1.89±0.24)(P<0.05).Compared to the NC group,cell proliferation activity decreased in the OE-LINC00641 group(P<0.05).Conclusion LINC00641 is downregulated in o-varian serous carcinoma and has an inhibitory effect on tumour cell proliferation.
