An in Vitro Screening Experiment Prepared with Liquid Oral Cellulose Gastric Ultrasound Developer

Abstract:Objective To prepare a convenient,liquid-to-use cellulose-based oral gastric ultrasound developer.Methods In this study,a large number of in vitro screening experiments were conducted to screen methyl cellulose,ethyl cellulose,carboxymethyl cellulose,hydroxypropyl methyl cellulose,polyglucose,gum Arabic,microcrystalline cellulose,colloidal microcrystalline cellulose and other materials.Screening experiments were conducted to investigate the degree of difficulty of brewing in different temperature wa-ter(cold water 10 degrees Celsius,warm water 40 degrees Celsius and hot water 90 degrees Celsius),and the quality of ultrasonic imaging after mixing.In order to obtain the best screening results,a comparative study was conducted using the currently used com-mercially available products.The ultrasonic imaging effects of the above common cellulose substances after single and complex water suspension were investigated,including the ultrasonic imaging effects of different concentrations and sizes of cellulose.Other evalua-tion tests include sedimentation volume ratio determination,centrifugal stability determination,freeze-thaw cycle stability test,water dilution test and so on.Results According to the above screening experiments,it was found that the selection of 2%microcrystalline cellulose(preferably with a particle size of 80~110 μm)had a good ultrasonic imaging effect,and the colloidal microcrystalline cel-lulose Avicel CL-611(with a concentration of 2.0%)had an excellent suspension effect on it.Therefore,micrystalline cellulose(preferably with a particle size of 80~110 μm)and colloidal micrystalline cellulose are ideal as ultrasonic imaging particles and sus-pension thickening agents of the gastric ultrasonic developer of the invention,respectively.The physical properties of the ideal formula-tion of oral gastric ultrasound developer are as follows:the sedimentation volume ratio is 1.0,the centrifugation is stable without strat-ification,the freeze-thaw cycle is stable without stratification,and the mixing time with water dilution is 6 seconds.Conclusion Through a large number of screening experiments,this study provides a convenient formulation of liquid ready-to-use oral gastric ul-trasound developer and its preparation method.
