Expression of Glucose Transporter 1 in Colon Cancer Cells and its Effect on Glucose Uptake

Abstract:Objective To study the expression of glucose transporter 1 ( Glut 1 ) in colon cancer cell lines and clinical samples and to explore the correlation between colon cancer progression and the expression. Methods The Glut1 protein expression in colon cancer tissues and cell lines was determined with Western blotting and immunohistochemisty respectively. The consumption of glucose in colon cancer cells was examined with Glucose ( GO) Assay Kit. Results Western blotting results showed that the expression of Glut1 in colon cancer cell lines significantly increased compared with that in tumor-adjacent tissues and in normal intestinal mucosa cells (P<0. 05). As immunohistochemisty results showed, the expression of Glut1 in cancer tissues increased remarkably compared with that in tumor-adjacent tissues (P<0. 05), and the expression was closely related to the pathological stage of colon cancer (P<0. 05). The results of Glucose (GO) Assay Kit suggested that the content of glucose in the culture media of colon cancer cell lines de-creased significantly compared with that in normal intestinal mucosa cells. Conclusion Glut1 expression is positively related to colon cancer progression.
