Morphology Observation of Dioleoyl Phosphatidyl Choline Combined with Poly ε-caprolactone Electrospun Fibers

Abstract:Objective To prepare poly ε-caprolactone electrospun fibers with homogeneous diameters using phosphalipid as an additive. Methods poly ε-caprolactone and dioleoyl phosphatidyl choline were dissolved in mixed solution of dichloromethane and dimethylformamide of 80 ﹕ 20 volume ratio forming homogeneous solution for electrospinning. Scanning electronic microscope was used to observe the fiber morphology. Wide angle X-ray diffraction was used to analyze the combination of dioleoyl phosphatidyl choline with poly ε-caprolactone. Fibers were immersed in the water to observe the dissolution of dioleoyl phosphatidyl. Results When the ratio of poly ε-caprolactone and dioleoyl phosphatidyl choline was 10 ﹕ 0 and 9 ﹕ 1, the sample presented beads-fibers hybrids. When the ra-tio was 8 ﹕ 2, the sample began to present the fiber structure, but showed adhesion between fibers. When the ratio was 7 ﹕ 3 and 6 ﹕4, fiber structure became clear and the diameters of fibers became homogeneous. When the ratio was 5 ﹕ 5, the fibers became thicker and the homogeneity became undesirable. When the ratio was 4 ﹕ 6 and 3 ﹕ 7, the beads-fibers hybrid structure presented again. When the ratio was 2 ﹕ 8, 1 ﹕ 9 and 0 ﹕ 10, there were only block mass structure. Wide angle X-ray diffraction analysis indicated that as the amount of dioleoyl phosphatidyl choline increased, the characteristic peaks of poly ε-caprolactone became weaker till they disappeared, which illustrated that the two materials could amalgamate together. When the ratio was 3 ﹕ 7, 2 ﹕ 8, 1 ﹕ 9 and 0 ﹕ 10, phospholipid could be dissolved from the samples. Conclusion Fibers with rather homogeneous diameter could be obtained by electro-spinning homogeneous solution by using dioleoyl phosphatidyl choline as an additive under appropriate ratio. And the combined fibers had no phospholipid dissolving out.
