Experimental Study on Differentiation of HPDMSCs into Cardiomyocyte-like Cells Induced by Salvianolic Acid B

Abstract：Objective To investigate the effect of salvianolic acid B on the differentiation of hpDMSCs into cardiomyocyte-like cells. Methods HpDMSCs were cultured in different groups:10% DMED culture group ( negative control group) , 10μmol/L 5-aza inducing group ( positive control group) , and 10 mg/L salvianolic acid B inducing group. Cell proliferation in the three groups were measured with MTS assay every 4 days. After three weeks, the expression of GATA4, ANF and cTnI was then detected with real-time pCR and Western blot. Results Compared with the negative control group, the cell proliferation rate of the two inducing groups sig-nificantly decreased, while the expression of GATA4, ANF and cTnI remarkably increased, especially in 10 mg/L salvianolic acid B inducing group. Conclusion Salvianolic acid B can induce the differentiation of hpDMSCs into cardiomyocyte-like cells, which has a better effect than 5-aza.
