Effects of Cdc42 Small Interfering RNA onP roliferation of Breast Cancer MCF7 Cells

Abstract:Objective To observe the effect of cell division cycle 42 (Cdc42) small interfering RNA (siRNA) on the prolifera-tion of breast cancer MCF7 cells.Methods The small interfering RNA in the targeted Cdc42 coding section was designed and synthe-sized, and transiently transfected into MCF7 cells.After 48 hours, the expressions of Cdc42 mRNA and protein after the transfection were detected with RT-PCR and Western blot respectively.The CCK8 as well as density experiments were applied to test the effect of Cdc42 siRNA on the reproductive capability of MCF7 cells.Results Both RT-PCR and Western blot showed that Cdc42 siRNA could notably lower the Cdc42 mRNA and its protein expression level in MCF7 cells.The proliferation of MCF7 cells was significantly inhibi-ted after the transfection of Cdc42 siRNA.Conclusion Cdc42 siRNA can significantly inhibit the proliferation of MCF7 cells, indica-ting that Cdc42 can be a promising target spot for treating breast cancer in the future.
