Influence of DPP-4 inhibitor and Valsartan on the Expression of CTGF in Diabetic Kidney Tissue

Abstract:Objective To investigate the protection mechanism of DPP-4 inhibitor and valsartan on diabetic nephropathy. Meth-ods Streptozotocin was used to establish mouse models of diabetic nephropathy on which intragastric administration was conducted with DPP-4 inhibitor, vildagliptin, and valsartan respectively. After 12-week treatment, the blood sugar and 24 h urine protein quantita-tive of each group were tested, and the CTGF in renal cortex was detected with immunohistochemistry. Results Staining in kidney is-sue of DN group was extensive and concentrated, especially in glomerular, and that in renal tubule and interstitial also enhanced. In vildagliptin treatment group and valsartan treatment group, the coloring range was similar, but the degree was significantly less than that of DN group (P<0. 01), with no significant difference between the two groups. There was difference between the combined treat-ment group and the two groups mentioned above ( P<0. 01 ) , and the CTGF level of the former group was lower than that of the DM group. Conclusion DPP-4 inhibitor, vildagliptin, and valsartan can reduce the damage to the kidney of mice with diabetes to a cer-tain extent, and the combined treatment is more effective.
