Study on the effect of glutathione in improving cognitive function,inflammatory oxidative stress and neurofactors in general anesthesia patients

Abstract:Objective To investigate the effects of glutathione treatment applied to general anesthesia patients on cognitive function and changes in inflammatory oxidative stress and neurofactors.Methods This study was a prospective randomized controlled study.120 cases of patients who were to receive sevoflurane inhalational anesthesia in Xingtai Central Hospital from December 2022 to March 2024 were selected as the study subjects,and were divided into the control group(60 patients)and the glutathione tablet group(60 patients)according to the method of randomized numerical table.Both groups of patients received sevoflurane inhalation anesthesia.The control group received routine treatment 2 weeks before anesthesia,while the glutathione tablet group received glutathione tablets in addition to the control group.The treatment lasted for 2 weeks and was observed until discharge.Cognitive function preoperatively and at 1 and 3 days postoperatively,inflammation-related indices,oxidative stress indices,and neurofactors preoperatively and at 1 day postoperatively,and adverse reactions during the observation period were compared between the 2 groups.Results At 1 and 3 d postoperatively,the scores of the brief mental status examination scale(MMSE)in the glutathione tablet group were(26.78±0.15)points and(27.91±0.32)points,respectively,which were higher than those of the control group(25.12±0.09)points and(27.49±0.60)points(t=73.506,4.784,P<0.05).At 1 d postoperatively,serum interleukin-1β(IL-1β),IL-6,tumor necrosis factor-α(TNF-α),C-reactive protein(CRP),malondialdehyde(MDA),cortisol(Cor),myelin basic protein(MBP),and neuron specific enolase(NSE)levels in the two groups were(47.97±6.63)ng/mL,(10.34±2.76)ng/L,(30.43±5.52)μg/L,(44.71±5.58)mg/L,(6.97±2.24)μmol/L,(215.44±22.67)mmol/L,(10.15±1.68)μg/L,and(23.35±3.09)μg/L,respectively,which were lower than those of the control group(54.12±8.54)ng/ml,(24.54±4.78)ng/L,(44.78±6.93)μg/L,(57.63±7.79)mg/L,(8.10±2.55)μmol/L,(291.33±31.45)mmol/L,(15.32±1.90)μg/L,(35.66±4.13)μg/L(t=4.406,19.928,12.546,10.444,2.579,15.163,15.790,18.486,P<0.05);serum glutathione peroxidase(GSH-Px),superoxide dismutase(SOD),nerve growth factor(NGF),and brain-derived neurotrophic factor(BDNF)levels were(55.29±3.45)U/ml,(65.23±4.71)U/mL,(663.44±70.22)ng/L,and(6.21±1.45)ng/L,respectively,which were higher than those in the control group of(41.54±3.19)U/ml,(50.21±3.52)U/ml,(576.32±(63.42)ng/L,(5.02±0.94)ng/L(t=22.667,19.786,7.132,5.334,P<0.05).Conclusion Glutathione could reduce sevoflurane inhalation anesthesia patients' body inflammation and oxidative stress,regulate the level of patients' body neurofactors,reduce nerve damage,and improve patients' cognitive function,and had good safety.
