The relationship between ATP5A1 and PDGFB expression in gastric cancer tissues and the long-term prognosis of patients after radical gastrectomy

Abstract:Objective To investigate the correlation between the expression levels of ATP synthase α-subunit(ATP5A1)and platelet-derived growth factor B(PDGFB)in gastric cancer tissues and the long-term prognosis of patients undergoing radical gastrectomy and their combined predictive value.Methods A total of 250 patients with gastric cancer who underwent radical gastrectomy in Feixian People's Hospital from March 2017 to March 2021 were selected as the study subjects,and were divided into survival group and death group according to the 3-year prognosis.The expression of ATP5A1 and PDGFB was detected by immunohistochemical staining.Univariate analysis and multivariable Logistic regression analysis were used to analyze the related factors of poor long-term prognosis in patients undergoing radical gastrectomy.The area under the curve(AUC)of the receiver operating characteristic(ROC)curve was used to analyze the predictive value of ATP5A1 and PDGFB single and combined detection for long-term poor prognosis in patients undergoing radical gastrectomy.Results After 3 years of follow-up,there were 83 cases(33.20%)in the survival group and 167 cases(66.80%)in the death group.The positive expression rates of ATP5A1 and PDGFB in the death group were significantly higher than those in the survival group(x2=38.716,P<0.001;x2=47.037,P<0.001)).The expression levels of ATP5A1 and PDGFB in gastric cancer tissues of patients with radical gastrectomy were correlated with tumor maximum diameter,degree of differentiation,depth of invasion and lymph node metastasis(P<0.05),but not with age,gender,carcinoembryonic antigen(CEA)and carbohydrate antigen(CA199)levels(P>0.05).Multivariate Logistic regression analysis showed that tumor diameter ≥ 5 cm,depth of invasion T3+T4,recurrence and metastasis,low degree of differentiation,high expression of ATP5A1 and high expression of PDGFB were independent risk factors for poor long-term prognosis in patients with radical gastrectomy(P<0.05).The AUC of combined detection of ATP5A1 and PDGFB in the long-term prognosis of patients with radical gastrectomy was 0.935(95%CI:0.903~0.967),which was significantly higher than that of single detection of ATP5A1 and PDGFB[0.869(95%CI:0.824~0.914),0.842(95%CI:0.792~0.892)],and the differences were statistically significant(Z=3.557,4.716,P<0.001).Conclusion The combined prediction of ATP5A1 and PDGFB expression levels in gastric cancer tissues demonstrates superior predictive value for long-term poor prognosis in patients undergoing radical gastrectomy,and provides an effective tool for assessing the prognosis of patients.
