The relationship between the expression of miR-20a and miR-122 and the count of HIV-1 DNA,HIV-1 RNA and CD4+ T lymphocytes in peripheral blood mononuclear cells of patients infected with HIV-1

Abstract:Objective The expression levels of micrornas(miR-20a)and miR-122 in peripheral blood mononuclear cells(PBMCs)infected with human immunodeficiency virus 1(HIV-1)were detected to investigate their relationship with HIV-1 DNA,HIV-1 RNA and CD4+ T lymphocyte count.Methods A total of 110 HIV-1 infected patients admitted to Nantong third people's hospital from May 2021 to January 2023 were selected as the HIV-1 group.According to HIV-1 DNA,they were divided into HIV-1 DNA high expression group(DHG group,65 cases)and HIV-1 DNA low expression group(DLG group,45 cases).According to HIV-1 RNA,they were divided into HIV-1 RNA high expression group(RHG group,49 cases)and HIV-1 RNA low expression group(RLG group,61 cases).According to the CD4+ T lymphocyte count,they were grouped into<200 cells/μl group(36 cases),200~350 cells/μl group(48 cases),and>350 cells/μl group(26 cases).Another 110 healthy subjects in the same period were selected as the control group.Fluorescence quantitative PCR was applied to detect PBMCs miR-20a,miR-122,HIV-1 DNA,and plasma HIV-1 RNA,the CD4+ T lymphocyte count was obtained by flow cytometry.The differences in the levels of miR-20a and miR-122 in PBMCs of patients in each group were compared.Pearson method was applied to analyze the correlation between the two indicators.Results The levels of miR-20a and miR-122 in PBMCs of HIV-1 group were higher than those of control group[(1.72±0.45)vs.(1.09±0.12),(1.51±0.38)vs.(1.04±0.07)](t=14.188,12.757,P<0.05).Compared with DLG group,the levels of miR-20a and miR-122 in PBMCs of HIV-1 group in DHG group were obviously higher[(1.91±0.48)vs.(1.45±0.41),(1.64±0.39)vs.(1.32±0.37)](t=5.239,4.320,P<0.05).The levels of miR-20a and miR-122 in PBMCs of HIV-1 group in RHG group were higher than those in RLG group[(1.85±0.46)vs.(1.62±0.44),(1.70±0.40)vs.(1.36±0.36)](t=2.670,4.685,P<0.05).The levels of miR-20a and miR-122 in the groups of<200 cells/μl,200~350 cells/μl and>350 cells/μl decreased gradually[(2.04±0.55),(1.67±0.46),(1.37±0.29),(1.73±0.40),(1.51±0.39),(1.21±0.33)](F=16.525,14.117,P<0.05).The correlation result showed that there was a positive correlation between miR-20a and miR-122 in PBMCs of HIV-1 group(r=0.651,P<0.05),both miR-20a and miR-122 had a obvious positive correlation with HIV-1 DNA(log2 DNA)(r=0.569,0.471,P<0.05),and a obvious positive correlation with HIV-1 RNA(log2 RNA)(r= 0.491,0.444,P<0.05),but a negative correlation with CD4+ T lymphocyte count(r=-0.555,-0.536,P<0.05).Conclusion The expression levels of miR-20a and miR-122 in PBMCs of HIV-1 infected individuals are elevated,they are related to the CD4+ T lymphocyte count and the quantitative detection result of HIV-1 virus DNA and RNA,and may be related to the establishment of a virus repository.Detecting both levels can reflect the patient's infection status.
