Abstract:Objective To study the effects and underlying mechanisms of miR-29a on the proliferation and apoptosis of placental trophoblast cells. Methods The expression of miR-29a and MCL1 mRNA was measured by RT-PCR. The effect of miR-29a on the proliferation of human trophoblast cells was measured by the CKK-8 assay.Flow cytometry was used to detect the effect of miR-29a on apoptosis. The protein levels of were detected by western blot. Results The proliferation of human extravillous trophoblast cells was decreased after overexpression of miR-29a. Apoptosis of human extravillous trophoblast cells was upregulated with the overexpression of miR-29a.The results of real-time PCR and western blots showed that overexpression of miR-29a inhibited MCL1 mRNA and protein levels. After interference with miR-29a,human villous trophoblast cell proliferation increased, the level of apoptosis decreased, and MCL1 mRNA and protein levels increased. Conclusion miR-29a inhibite the proliferation of trophoblasts outside of human villi,down-regulate protein levels of MCL1 and promot apoptosis in trophoblasts.
