Expression of chemokine receptors CCR1 and CCR5 in gastric carcinoma and its clinical significance
Abstract:Objective To investigate the expression and distribution of chemokine receptors CCR1 and CCR5 in gastric carcinoma and adjacent normal tissues,to analyze their correlation with clinicopathological features,and to investigate whether the expression of these two factors in gastric cancer tissues correlate. Methods The expression of CCR1 and CCR5 in gastric cancer tissues and adjacent normal tissues was detected by immunohistochemical methods and by western blot. The purpose was to record and analyze expression levels,to find the correlation between these and pathological characteristics,and to analyze the correlation. Results Immunohistochemistry showed that the positive rate of CCR1 in gastric cancer tissues was 52%(31/60).The positive rate of CCR1 in normal tissues was 25%(15/60). The positive rate of CCR1 in gastric cancer tissues was significantly higher than that of adjacent normal tissues(P < 0.05). The positive rate of CCR5 in gastric cancer tissues was 60%,and the positive rate of CCR5 in adjacent normal tissues was 28%. The positive rate of CCR5 in the two tissues was very different and statistically significant(P <0.05).Western blot results showed that the relative expression levels of CCR1 and CCR5 in gastric cancer tissues were 2.134 ± 0.193 and 2.736 ± 0.125, respectively,but the relative expression levels of CCR1 and CCR5 were 1.378 ± 0.047 and 0.971 ± 0.027 in normal tissues, respectively(P < 0.05). The expression level of CCR1 was not correlated with gender,age,differentiation degree or depth of invasion(P>0.05),but it was correlated with tumor differentiation and lymph node metastasis(P < 0.05). The expression of CCR5 in tissues was not correlated with sex,age and differentiation of gastric cancer(P > 0.05)but was positively correlated with invasion depth and lymph node metastasis(P < 0.05). CCR1 was positively correlated with CCR5 expression(P < 0.05). Conclusion The expression of CCR1 and CCR5 may be associated with the occurrence and development of gastric cancer. It may be helpful to diagnose gastric cancer using CCR1 and CCR5.
