Optimization of the Gametophyte Cultivation Condition of Undaria Pinnatifida

Abstract:The main effects of different temperatures (18,21,24,and 27℃),light intensities (50,80,110 and 140μE · m-1 · s-1),cultivation methods (static culture,magnetic stirring culture,inflatable suspension culture),and inoculation densities (0.1,0.2,0.4,0.6,0.8,and 1.0g/L) on the growth of Undaria gametophytes were studied under laboratory conditions.The results showed that the light intensity and the temperature as well as their interaction have great impacts on the gametophyte growth.The studies revealed that the optimum environmental factors for the growth of U.pinnatifida were 50 E m-1 s-1 and 21 ℃.From the 5th to the 10th day,the U.pinnatifida gametophytegrowth rate was significantly higher than that on other days,and consequently to maintain a high growth rate,the 10th day culture was chosen for subculture.The cultivation method and the inoculation density as well as their interaction had a great impact on the growth of gametophytes.The results showed thatthe optimum factors for the growth of U.pinnatifida were 0.1-0.2g/L and inflatable suspension for the initial inoculation density andas the cultivation method,respectively.Generally speaking,U.pinnatifida grows well underthe following conditions:temperature 21 ℃,light intensity50μE · m-1 · s-1,inoculation density 0.1-0.2g/Land inflatable suspension cultivation.
