Screening of Serum Exercise-sensitive miRNA in Mice

Abstract:Objective To study the effect of different exercise intensity on the circulating miRNA in serum of mice. Methods C57BL/6 mice were randomly divided into control group(Con) and exercise groups: moderate-intensity exercise group(M) and high-intensity exercise group(H). Exercise groups were selected to perform treadmill exercise at a speed of 10 m/min and 20 m/min respectively for 30 minutes each time. Twentyfour hours after moderate-intensity exercise and high-intensity exercise were regarded as recovery group MR and HR, respectively. Real-time PCR was used to detect the changes of miR-1a, 26a, 133a and 146a in serum and soleus of mice. Results MiR-1a, 26a and 133a were significantly increased (P < 0.01) and miR-146a was significantly elevated (P < 0.05) in moderate-intensity exercise. The level of miR-1a and 133a in serum increased significantly during high-intensity exercise (P < 0.05). MiR-26a was significantly elevated (P < 0.01), and miR-146a had no significant changes. MiR-1a and 133a were significantly decreased in skeletal muscle during moderate exercise (P < 0.05), while miR-26a and 146a had a decreasing trend (P > 0.05). In high-intensity exercise, miR-1a and miR-133a in skeletal muscle of mice were significantly increased (P < 0.05, P < 0.01), and there were no significant change in miR-26a and 146a. After 24 h, the miRNA in serum and soleus recovered to the control group level. Conclusion The changes of serum circulating miRNA in mice were caused by exercise, and different exercise intensity had different effects on miRNA in serum. The soleus muscle may be one of its sources.
