Development and Validation of a Closed Cell Culture Plate for Space Applications

Abstract:Objective In order to make the cell culture plates fit for space experiments,to use reagents more efficiently in cell culture experiments,and to improve the anti-contamination ability of cell culture materials,a fully transparent closed cell culture plate with controlled inner flow path and anti-leakage/anti-contamination capability was proposed.Methods The arrangement of the flow path control bar which is the core structure of the culture plate was optimized based on the finite element simulation (FES).The optimal cell culture plate was fabricated,then the mid-long-term cell culture experiments were carried out in the cell culture loop with the aforementioned cell culture plate as its key component.Results The optimal cell culture plate with three longitudinally arranged bars could form well-controlled flow channels in its chamber.When the flow velocity at inlet and outlet was 1 mL/min,the fluid shear stress in the chamber was only 4.8 × 10-5Pa.And there was almost no flow dead zone.The cells cultured in the cell culture loop with the cell culture plate as its core component could proliferate normally throughout the 15 d experimental period.Conclusion The cell culture plate has the following advantages:the replacement of old and new cell culture reagents and residual gas discharge are more thorough;anti-leakage and anti-contamination capability are significantly improved;and reagent usage rate is increased.It is suitable for the mid-long-term culture of the typical cells used in the space medical biology experiments.
