Isolation of Differentially Expressed Stress Genes in Rat Heart Tissue after Repeated High +Gz Exposures
Abstract:Objective To isolate the specific stress genes in hearts from the rat repeated +Gz exposure model. Method Rats were exposed to +10 Gz three times on an animal centrifuge, each for 3 min with 30 min internal. Total RNA was isolated from rat hearts 1 h after exposures and control hearts. Poly A+ RNA was purified from total RNA. cDNA probe labeled with 32 P and hybridized with Atlas Rat Stress Array. Transcription of differentially expressed genes were corroborated by using semi-quantitative RT-PCR. Result Eight genes exhibited elevation in expression and 2 genes exhibited depression in expression at 1 h after repeated +Gz exposure. Genes for heat shock protein 70(HSP70), P21 and heme oxygenase-1(HO-1) were selected as candidates for relative specific genes. Upregulated expression of P21 and HO-1 and downregulated expression of HSP70 were corroborated by semi-quantitative RT-PCR. Conclusion Atlas Rat Stress Array can be used to determine information about stress genes in rat heart exposed to repeated +Gz.
