Effectiveness of Sintilimab Combined with Chemotherapy in Advanced Gastric Adenocarcinoma

Abstract:Objective To evaluate the efficacy of sintilimab combined with chemotherapy in advanced gastric adenocarcinoma and its impact on peripheral blood levels of signal transducer and activator of transcription-3(STAT-3),STAT-5 and interleukin(IL)-17A.Methods A total of 80 patients with advanced gastric adenocarcinoma admitted to Xinyang Hospital of Traditional Chinese Medicine from October 2021 to September 2023 were randomly divided into control group and observation group according to a random number table.The control group(40 cases)received conventional chemotherapy,whereas the observation group(40 cases)received sintilimab in combination with conventional chemotherapy,21 days for one cycle,and continued treatment for 6 cycles.The short-term efficacy of the two groups was compared.Peripheral blood STAT-3,STAT-5,IL-17A,tumor markers,quality of life,and adverse reactions prior to and following treatment were compared between the two groups.Results The observation group had higher objective response rate(ORR)and disease control rate(DCR)than the control group(P<0.05).After treatment,the expression levels of STAT-3,STAT-5,IL-17A,serum levels of glucose carbohydrate antigen 199(CA199),carcinoembryonic antigen(CEA),and tumor supplied group of factors(TSGF)were reduced relative to pretreatment values in both groups,with the observation group showing lower levels than the control group(P<0.05).After treatment,the score on the European organisation for research and treatment of cancer quality of life questionnaire-stomach cancer module(EORTC QLQ-STO22)was higher in the observation group than in the control group(P<0.05).The incidence of adverse reactions did not differ significantly between the two groups(P>0.05).Conclusion Combined sindilitumab in the treatment of advanced gastric adenocarcinoma can effectively improve the efficacy,regulate the expression levels of STAT-3,STAT-5 and IL-17A,tumor marker levels can be reduced and patients' quality of life improved,without an increase in adverse reactions.
