Comparison of the Efficacy and Safety of Combination Chemotherapy with Tislelizumab and Sintilimab in the Treatment of Advanced Non-small Cell Lung Cancer

Abstract:Objective To evaluate the differences in short-term efficacy and safety between tislelizumab and sintilimab combined with chemotherapy in advanced non-small cell lung cancer(NSCLC).Methods A total of 90 patients with advanced NSCLC and negative driver genes who were treated at the Second Outpatient Department of Henan Province from January 2021 to April 2023 were enrolled.They were randomly divided into the tislelizumab plus chemotherapy group(control group,45 cases)and the sintilimab plus chemotherapy group(observation group,45 cases)using a random number table method.Both groups received the same chemotherapy regimen of paclitaxel(135 mg·m-2)combined with cisplatin(30 mg·m-2).Immunotherapy was administered via intravenous infusion with tislelizumab(200 mg)or sintilimab(200 mg),with a 21-day cycle repeated for 3 cycles.The primary endpoint was the disease control rate(DCR),while secondary endpoints included tumor marker levels,immune function indicators,total adverse reaction incidence,and 1-year survival rate.Results There was no statistically significant difference in disease control rates between the two groups(P>0.05).Compared to pre-treatment levels,tumor marker levels decreased in both groups post-treatment;however,no statistically significant difference was observed in serum tumor marker levels between the two groups post-treatment(P>0.05).Relative to pre-treatment,post-treatment levels of CD3+,CD4+,and CD4+/CD8+increased in both groups,while CD8+levels decreased;no statistically significant differences were found in post-treatment levels of CD3+,CD4+,CD8+,or CD4+/CD8+between the two groups(P>0.05).The total incidence of adverse reactions was lower in the observation group than in the control group(P<0.05).Kaplan-Meier survival curve analysis revealed no statistically significant difference in 1-year survival rates between the two groups(P>0.05).Conclusion The short-term efficacy of tislelizumab or sintilimab combined with chemotherapy is similar for driver gene-negative advanced NSCLC.Compared to the tislelizumab plus chemotherapy regimen,sintilimab plus chemotherapy carries a lower risk of adverse reactions and demonstrates acceptable safety.
