Mechanistic Investigation of Alginate Oligosaccharides Alleviating ox-LDL-Induced Endothelial Cell Injury by Intervening in HMGB1-Regulated GSK-3β Signaling Pathway

Abstract:Objective To investigate the impact of alginate oligosaccharides(AOS)on atherosclerosis(AS)and its underlying potential mechanisms.Methods Human umbilical vein endothelial cells(HUVECs)were used as the research subjects.Cell viability was assessed using the CCK-8 assay.Gene expression was detected by qRT-PCR and Western blot.Cell apoptosis and reactive oxygen species levels were evaluated through flow cytometry.Superoxide dismutase(SOD)activity was measured using an SOD kit.The contents of inflammatory factors interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were determined by ELISA.Results AOS effectively alleviated the inhibitory effect of oxidized low-density lipoprotein(ox-LDL)on HUVECs viability and partially reversed the promoting effect of ox-LDL on high mobility group box 1 protein(HMGB1)expression.The administration of AOS significantly attenuated the ox-LDL-induced suppression of HUVECs viability,promotion of apoptosis,and exacerbation of oxidative stress and inflammatory response.However,HMGB1 overexpression partially counteracted these beneficial effects of AOS.In ox-LDL-induced HUVECs,AOS successfully activated the glycogen synthase kinase-3β(GSK-3β)pathway by inhibiting HMGB1 expression.Conclusion AOS demonstrated potential therapeutic effects on AS,and its mechanism might be related to the regulation of HMGB1 expression and activation of the GSK-3β pathway,providing a new perspective for AS treatment.
