Mendelian Randomization Study on the Association Between Statin Use and Lung Cancer Risk

Abstract:Objective This study explored the causality between statin use and lung cancer through Mendelian randomization(MR)analysis.Methods Genome-wide association study(GWAS)data of statin use and lung cancer were downloaded from Finnish and integrative epidemiology unit(IEU)open GWAS project database,respectively.MR analysis was performed using methods including inverse-variance weighting(IVW),weighted median,MR Egger,and weighted mode.The heterogeneity,pleiotropy,and sensitivity analysis were also conducted.Results IVW analysis observed a causal association between statin use and lung cancer(OR=0.88,95%CI:0.78-1.00,P=0.042),small cell lung cancer(OR=0.90,95%CI:0.82-0.99,P=0.029),and lung adenocarcinoma(OR=0.94,95%CI:0.90-0.99,P=0.016).Taking statins can reduce the risk of developing lung cancer,small cell lung cancer,and adenocarcinoma of the lung.No causal relationship was found between statin use and lung squamous cell carcinoma(OR=0.95,95%CI:0.89-1.02,P=0.158).Conclusion The use of statins may reduce the risk of lung cancer,potentially having a significant impact on the prevention and treatment of lung cancer.
