Role of Circadian Rhythm Genes in the Prognosis of Colorectal Cancer and the Construction of a Prediction Model

Abstract:Objective To identify circadian rhythm genes associated with the prognosis of colorectal cancer and construct a prognostic prediction model based on bioinformatics analysis methods.Methods The messenger ribonucleic acid(mRNA)sequencing data and clinical information from 647 colorectal cancer samples and 51 adjacent tissue samples were obtained from The Cancer Genome Atlas(TCGA)database.The mRNA expression profile data were analyzed using the R language software to screen for differentially expressed genes(DEGs).By intersecting DEGs with 2 091 circadian rhythm-related genes from the GeneCards database,76 circadian rhythm-related DEGs were identified.Univariate Cox regression and LASSO regression analyses were performed to screen for five key genes,which were then subjected to multivariate Cox regression analysis to construct a prognostic model based on these key genes.Results Multivariate analysis revealed that AMH,COMP,SIX1,HTR2C,and MMP1 were independent predictors of overall survival(OS)in colorectal cancer patients(P<0.05).The nomogram model indicated that high expression of MMP1 was positively correlated with extended OS,while high expression of AMH,COMP,SIX1,and HTR2C was significantly associated with reduced OS.The time-dependent receiver operating characteristic(ROC)curve showed that the area under the curve(AUC)values of the model for predicting 1-year,3-year,and 5-year OS were 0.607,0.652,and 0.718,respectively.Conclusion A prognostic model based on circadian rhythm-related genes offers a novel tool for survival prediction in colorectal cancer patients.
