Relationship and Meaning of ARID1A Expression and Stromal Infiltrating Lymphocytes in Colorectal Adenocarcinoma

Abstract：Objective To investigate the expression of adenine-thymine(AT)rich binding domain containing protein 1A(ARID1A)protein in colorectal adenocarcinoma and its relationship with interstitial infiltrating lymphocytes(CD4+TILs,CD8+TILs).Methods A retrospective analysis was performed for 115 radical colorectal cancer specimens at the Fifth Clinical Medical College,Henan University of Traditional Chinese Medicine(Zhengzhou People's Hospital)from January 2019 to December 2022,and immunohistochemistry Envision was used to detect the loss of ARID1A and the degree of interstitial infiltrating lymphocytes(CD4+TILs,CD8+TILs).Results In colorectal adenocarcinoma,ARID1A was expressed in 90 cases(78.26％)and absent in 25 cases(21.73％),which was higher than that of adjacent normal tissues(P＜0.05).Compared with the ARID1A expression group,the infiltration density of CD4+TILs and CD8+TILs in the deletion group decreased significantly(P＜0.05).At the same time,it was also found that the infiltration density of CD4+TILs and CD8+TILs in the tumor center was lower than that at the tumor margin.There was a significant difference between the deletion group of ARID1A and lymph node metastasis,pTNM and histological grade(P＜0.01),but not significant difference with the age,gender,tumor diameter and lesion location of the patients(P＞0.05).ARID1A deletion was positively correlated with CD4+TILs(rs=0.264,P＜0.01)and CD8+TILs(rs=0.213,P＜0.01)in colorectal adenocarcinoma.Conclusion In colorectal adenocarcinoma,ARID1A deletion is associated with decreased infiltration of interstitial infiltrating lymphocytes(CD4+TILs and CD8+TILs),which provides reliable experimental data and theoretical basis for immunotherapy in patients.
