In Vitro Real-Time Quantitative Observation Method of Apoptosis Induced by Nanosecond Pulsed Electric Fields

Abstract:Objective This study aims to explore the apoptosis-inducing biological effects of nanosecond pulsed electric fields on tumor cells and identify suitable research methods for observing and evaluating the dose-effect relationship in primary hospitals.Methods A cell model was established under in vitro culture conditions,and nanosecond pulsed electric field therapy was applied externally.Apoptosis induction was measured using fluorescence observation,with changes in fluorescence recorded before and after treatment.Electric field parameters were optimized,and morphological changes and quantitative analysis of apoptosis were conducted.Results Nanosecond pulse-induced apoptosis exhibited a dose-effect relationship.Under electric field parameters of 30 000 V·cm-1 field strength,300 ns pulse width,and 1 Hz frequency,cellular responses to the external nanosecond pulsed electric field showed progressive stability at 1,2,and 3 hours post-treatment.Conclusion The in vitro assay facilitates intuitive quantitative assessment of the electromagnetic biological effects of nanosecond pulsed electric fields for clinicians.
