Clinical Relationship Between NLRP3 Gene Polymorphism and Susceptibility to Bronchial Asthma in Children

Abstract:Objective To explore the clinical relationship between nucleotide-binding and oligomerization domain(NOD)-like receptor family pyrin domain containing 3(NLRP3)gene polymorphism and susceptibility to bronchial asthma in children.Methods A total of 104 children with bronchial asthma admitted to the Third People's Hospital of Henan Province from February 2020 to January 2023 were selected as the asthma group,and 104 healthy children with normal physical examination results were included in the control group using propensity score matching method and matched with gender,age,etc 1∶1.The NLRP3 gene polymorphism between the two group were compared,and the clinical relationship between it and susceptibility to bronchial asthma in children was analyzed.Results There were no differences in gender,age,body mass index(BMI),premature birth,passive smoking and breastfeeding between the asthma group and the control group(P>0.05),but the proportion of asthma family history,interleukin-1β(IL-1 β)and interleukin-18(IL-18)in the asthma group were higher than those in the control group(P<0.05),while the forced expiratory volume in first second(FEV1),forced expiratory volume in first second/forced vital capacity(FEV1/FVC)and peak expiratory flow(PEF)were lower than those in the control group(P<0.05).The GG genotype and allele G frequency at the rs10754558 locus of the NLRP3 gene in the asthma group were higher than those in the control group,while the allele C frequency was lower than that in the control group(P<0.05).Logistic regression analysis showed that the GG genotype(OR=2.954)and allele G(OR=3.297)at the rs10754558 locus of the NLRP3 gene were risk factors for susceptibility to bronchial asthma in children(P<0.05).The children with G and GG genotypes in the proportion of asthma family history,IL-1β and IL-18 were higher than those with CC genotype,and the children with GG genotype was highest,while FEV1,FEV1/FVC and PEF were lower than those with CC genotype,and the children with GG genotype was lowest(P<0.05).Conclusion The GG genotype and allele G at the rs10754558 locus of the NLRP3 gene are associated with susceptibility to bronchial asthma in children.
