Effect of Oncostatin M on Endometrial Cancer Cells Proliferation and Apoptosis

Abstract:Objective To investigate the effects of tumor oncostatin M(OSM)on the proliferation,apoptosis and cycle of endometrial cancer cells.Methods Endometrial cancer cell lines HEC-1B and Ishikawa were treated with different concentrations of recombinant human tumor oncostain M(rhOSM)(0,25,50,100 μg·L-1),and the expressions of OSM and tumor suppressor receptor(OSMR)were detected by Western blot.The appropriate concentration(50 μg·L-1)was selected for subsequent experiments.The experiment was divided into two groups:the control group without rhOSM(0 μg·L-1)treatment and the experimental group with rhOSM(50 μg·L-1)treatment.Cell proliferation was detected by CCK-8 and plate cloning.Cell migration was determined by scratch assay.The changes of membrane potential were detected by JC-1.Western blot and RT-qPCR were used to detect apoptosis and cell cycle related proteins.Results Compared with the control group,the proliferation and cloning ability of the experimental group were decreased.Cell migration ability was significantly reduced.The mitochondrial membrane potential was decreased.The expression of proapoptosis-related protein Caspase-3 was increased,the ratio of Bax/Bcl-2 was up-regulated.The expression of cycle-associated protein P53 was up-regulated,CDK4 expression was down-regulated,and p-STAT3 was activated(P<0.05).Conclusion OSM inhibits cell cycle and induces cell apoptosis by affecting cell apoptosis and cell cycle related gene protein expression.
