Effect of Ketorolac Tromethamine on Mechanical Ventilation-Induced Lung Injury in Mice

Abstract:Objective To investigate the potential role of the nonsteroidal anti-inflammatory drug ketorolac tromethamine(KT)in the process of ventilator-induced lung injury(VILI)caused by mechanical ventilation(MV).Methods Mouse lung injury model was established by MV.The lung tissue was taken for hematoxylin and eosin(HE)staining,and the wet-to-dry weight ratio of the lung tissue was measured.Bronchoalveolar lavage fluid was collected and analyzed for lactate dehydrogenase(LDH)activity,total protein concentration,and neutrophil count.The levels of multiple inflammatory cytokines in bronchoalveolar lavage fluid were measured using enzyme-linked immunosorbent assay.Results MV induced severe acute injury in murine pulmonary tissue,characterized by marked edema and substantial granulocyte infiltration.After KT,the HE score decreased,and the pulmonary edema caused by MV in mice decreased in a dose-dependent manner.The levels of LDH,total protein,and various inflammatory factors in mice bronchoalveolar lavage fluid increased,and the number of neutrophils increased.After KT administration,LDH levels,total protein levels,neutrophil count,and inflammatory cytokine levels decreased.Conclusion KT reduces the lung injury caused by MV in mice,reduces the number of neutrophils in bronchoalveolar lavage fluid and the production of pro-inflammatory mediators.
