Expression of Hypoxia Inducible Factor-1α and Four and a Half LIM Domain 1 in Model Rats of Persistent Pulmonary Hypertension of the New Born

Abstract:Objective To explore the changes of hypoxia inducible factor-1α(Hif1α)and four and a half LIM domain protein 1(Fhl1)expression levels in lung tissues of model rats of persistent pulmonary hypertension of the new born(PPHN).Methods Six pregnant rats were randomly divided into control group(3 cases)and PPHN group(3 cases).On the 19th day of pregnancy,the PPHN group was given low oxygen(fraction of inspired oxygen was 10%-12%)in combination with piperine to establish a PPHN rat model,the control group was given normal oxygen concentrations(21%oxygen absorption).The fetuses were born by cesarean section on day 22 of gestation.Blood,lung tissues and hearts were collected and examined.Parameters including plasma brain natriuretic peptide(BNP)concentration,the right ventricular hypertrophy index,the percentage of medial wall thickness(MT%),the percentage of medial wall area(MA%)were used to estimate the development of PPHN.The protein expression level of Hif1α and Fhl1 were detected by Western blot.Results Compared to the control group,pulmonary small artery wall became thicker,the vascular cavity was narrower,pulmonary small artery MT%,MA%,right ventricular thickness index,plasma BNP concentration increased in the PPHN model group(P<0.01).PPHN model group Hif1α,Fhl1 protein expression levels were higher in the PPHN model group(P<0.05).Conclusion The expression of Hif1αand Fhl1 protein in lung tissue in PPHN model rats was up-regulated,suggesting that Hif1α and Fhl1 may be closely related to the pathogenesis of PPHN.
