Effects of Aspirin Combined with Low Molecular Weight Heparin on Neurological Impairment in Rats with Delayed Encephalopathy Caused by Carbon Monoxide Poisoning

Abstract:Objective To investigate the effects of aspirin combined with low molecular weight heparin on nerve function impairment in rats with delayed encephalopathy after acute carbon monoxide poisoning(DEACMP).Methods The DEACMP model was prepared by intraperitoneal injection of pure carbon monoxide gas.The rats with successful modeling were randomly divided into model group,hyperbaric oxygen group for 30 days,hyperbaric oxygen group for 14 days,hyperbaric oxygen group for 14 days plus aspirin combined with low molecular weight heparin group,and 10 rats without modeling were set as normal group.Rats in the 30-day hyperbaric oxygen group and 14-day hyperbaric oxygen group were treated with hyperbaric oxygen for 30 days and 14 days,respectively.Rats in the 14-day hyperbaric oxygen+aspirin combined with low molecular weight heparin group were given 9 mg·kg-1aspirin intragastric and 3 U·mL-1 low molecular weight heparin subcutaneous injection during hyperbaric oxygen treatment for 30 days.The spatial memory ability of rats was determined by Morris water maze test,the level of malondialdehyde(MDA)and superoxide dismutase(SOD)in serum of rats were determined by ELISA,the prothrombin time(PT)and thrombin time(TT)of rats were monitored by automatic coagulation analyzer,and the brain tissue injury of rats was observed by HE staining.The apoptosis rate of rat hippocampal tissue was observed by TUNEL staining.Results Compared with normal group,the escape latency of rats in model group was prolonged,the number of crossing platforms was reduced,SOD level was decreased,MDA level was increased,PT and TT were shortened,and apoptosis rate was increased(P<0.05).Compared with model group,the escape latency of rats in 30-day hyperbaric oxygen group and 14-day hyperbaric oxygen group was shortened,the number of crossing platforms was increased,SOD activity was increased,MDA level was decreased,PT and TT were prolonged,and apoptosis rate was decreased(P<0.05).Compared with the 30-day hyperbaric oxygen group,the escape latency of rats in the 14-day hyperbaric oxygen group was shortened,the number of crossing platforms was increased,SOD activity was increased,MDA level was decreased,PT and TT were prolonged,and apoptosis rate was decreased(P<0.05).Compared with 14-day hyperbaric oxygen group,14-day hyperbaric oxygen+aspirin combined with low molecular weight heparin group,the escape latency was shortened,the number of crossing platforms was increased,SOD activity was increased,MDA level was decreased,PT and TT were prolonged,and apoptosis rate was decreased(P<0.05).Conclusion During hyperbaric oxygen short-term treatment of DEACMP,combined use of aspirin and low molecular weight heparin can significantly improve spatial memory ability,reduce oxidative damage and improve brain microcirculation.
