Effect of Histone Deacetylase 3 Inhibitor RGFP966 Combined with Cisplatin on Proliferation and Apoptosis of Cervical Cancer Cells

Abstract：Objective To explore the influence of histone deacetylase 3(HDAC3)inhibitor RGFP966 combined with cisplatin on the proliferation and apoptosis of cervical cancer cells.Methods Cervical cancer CaSki cells were divided into control group,RGFP966 group,cisplatin group,and RGFP966+cisplatin group and treated with HDAC3 inhibitor RGFP966 and cisplatin.Real time fluorescence quantitative PCR was used to detect HDAC3 gene in tissues and cells.Western blot was used to detect HDAC3 in tissues and cells and apoptotic protein Caspase-7,Caspase-3 and Caspase-9.Immunohistochemistry was used to detect HDAC3 protein in tissues,MTT and migration test were used to estimateproliferation of cervical cancer cells,Transwell was used to invasion assays,Annexin V-FITC/PI double staining were used to estimateapoptosis of cervical cancer cells,finally analyzed by SPSS 18.0 data statistics software.Results The effect of cisplatin on the relative survival rate of Hela and CaSki cells was dose-dependent,and the relative survival rate of 2 mg·L-1 Hela and CaSki cells was the lowest.The expression of HDAC3 in cervical cancer tissue was higher than that in adjacent tissues,both HDAC3 inhibitor RGFP966 and cisplatin could reduce the expression of HDAC3 gene and protein,but accelerated the expression of Caspase-7,Caspase-3 and Caspase-9,reduced proliferation and promoted apoptosis,the joint action of the two was more obvious.Conclusion HDAC3 inhibitor RGFP966 combined with cisplatin can significantly change the proliferation and apoptosis of cervical cancer cells,and the combination of the two drugs has a more significant effect on the treatment of cervical cancer.
