Changes and Relationship Between Gut Microbiota and Serum SIRT1,TLR4,NF-κB in Patients with Ulcerative Colitis

Abstract:Objective To explore the changes and relationship between gut microbiota and serum silencing information regulatory factor 2-related enzyme 1(SIRT1),Toll like receptor 4(TLR4),and nuclear factor κB(NF-κB)in patients with ulcerative colitis.Methods A total of 120 patients with ulcerative colitis who received treatment in the First Affiliated Hospital of Henan University of Medicine from January 2020 to June 2024 were selected as the observation group,and 120 normal patients who underwent physical examination during the same period were selected as the control group.The intestinal flora and serum levels of SIRT1,TLR4 and NF-κB in the two groups were compared,and the correlation between the intestinal flora and serum levels of SIRT1,TLR4 and NF-κB in the observation group was analyzed.Results The serum SIRT1 level in the observation group was lower than that in the control group,while the relative expression levels of TLR4 and NF-κB were higher than those in the control group(P<0.05).Pearson correlation analysis showed that SIRT1 levels were negatively correlated with the distribution of Escherichia coli,Enterococcus,and Lactobacillus,and positively correlated with the distribution of Bifidobacterium.The expression levels of TLR4 and NF-κB were positively correlated with the distribution of Escherichia coli,Enterococcus,and Lactobacillus,and negatively correlated with the distribution of Bifidobacterium.Receiver operating characteristic(ROC)curve analysis showed that serum SIRT1 level and relative expression of TLR4 and NF-κB had high diagnostic value for the occurrence of ulcerative colitis.The area under the curve(AUC)values were 0.931,0.976,and 0.997 respectively.Conclusion The serum levels of SIRT1 and the relative expression levels of TLR4 and NF-κB in patients with ulcerative colitis are significantly different from those in healthy individuals.SIRT1,TLR4,and NF-κB all have high diagnostic value for ulcerative colitis.
