Effects of Berberine on High Glucose Induced Injury in Mouse Renal MPC5 Podocytes

Abstract:Objective To investigate the effects of berberine on MPC5 injury of mouse renal podocyte induced by high glucose and its molecular mechanism.Methods MPC5 cells were cultured in vitro and divided into control group,model group,low-dose berberine group,medium-dose berberine group,high-dose berberine group,anti-miR-NC group,anti-miR-4270 group,high-dose berberine+miR-NC group,high-dose berberine+miR-4270 group.RT-qPCR was used to detect the expression level of miR-4270.Flow cytometry was used to detect cell apoptosis.Western blot was used to detect the expression of apoptosis-related proteins.ELISA was used to detect the expression levels of inflammatory factors.Results High glucose treatment promoted apoptosis and inflammatory response of MPC5 cells(P<0.05).Berberine treatment or miR-4270 downregulation inhibited apoptosis and inflammatory response of MPC5 cells(P<0.05).Berberine treatment reduced the expression level of miR-4270(P<0.05),and the up-regulation of miR-4270 expression reversed the effects of berberine on apoptosis and inflammatory response of MPC5 cells treated with high glucose(P<0.05).Conclusion Berberine alleviates high glucose induced MPC5 cell injury by down-regulating the expression of miR-4270.
